    Non-treated ACHN cells and IL-4 (HZ-1004) treated ACHN cells were subjected to SDS PAGE followed by western blot with 86641-1-RR (Phospho-STAT6 (Tyr641) antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Vinculin (66305-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 86641-1-PBS in a different storage buffer formulation.

Cells were fixed and permeabilized with Flow Cytometry Phosphorylated Protein Fixation/Permeabilization Kit (PF00026). This data was developed using the same antibody clone with 86641-1-PBS in a different storage buffer formulation.

背景介绍

STAT6, also named as Signal transducer and activator of transcription 6, is a 847 amino acid protein, which belongs to the transcription factor STAT family. p-STAT6(Tyr641) is a biomarker of Th2-high inflammation, targeted by IL-4Rα blockers (dupilumab) and JAK1/3 inhibitors. High p-STAT6 in TAMs correlates with M2 polarization and radioresistance, blocking IL-4 or JAK1/3 decreases Tyr641 phosphorylation and restores anti-tumor immunity. (PMID: 37474750)
