    Chromatin was prepared from HeLa cells. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of Phospho-POLR2A (Ser7) (83932-1-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR. This data was developed using the same antibody clone with 83932-1-PBS in a different storage buffer formulation.

    Non-treated HEK-293 cells, phosphatase inhibitor treated HEK-293 cells and λ phosphatase treated HEK-293 cells were subjected to SDS PAGE followed by western blot with 83932-1-RR (POLR2A antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83932-1-PBS in a different storage buffer formulation.

背景介绍

DNA-directed RNA polymerase II subunit RPB1(POLR2A), is part of the core element of the basic RNA polymerase II transcription mechanism. During transcription elongation, Pol II moves on the template as the transcript elongates. Elongation is influenced by the phosphorylation status of the C-terminal domain (CTD) of Pol II largest subunit (RPB1), which serves as a platform for the assembly of factors that regulate transcription initiation, elongation, termination, and mRNA processing.
