体外活性

4-AC是一种天然抗氧化剂,表现出强大的保护RPE细胞免受氧化应激引起的坏死能力。从机制上讲,4-AC通过阻断氧化应激引起的细胞ROS增加,并通过稳定及诱导NRF2转录因子的核内转移,上调NQO1和HO-1基因,从而发挥作用。进一步研究显示,NQO1、HO-1和NRF2是4-AC保护RPE细胞免遭tBHP引发死亡所必需的。t2,5-Di-tert-butylhydroquinone,作为NRF2的稳定剂,一致模拟了4-AC对抗tBHP引起的RPE细胞死亡的保护效果[1]。
