MG-63 cells were cultured in DMEM supplemented with 10% fetal bovine serum, 2.5 mM L-glutamine, 100 U/mL penicillin, and 100 μg/mL streptomycin sulfate. An aliquot of the cell culture supernatant was removed, assayed for CXCL6, and measured 1.3 ng/mL.

Human peripheral blood mononuclear cells (PBMC) were cultured unstimulated or stimulated with 10 μg/mL PHA for 3 days. The mean CXCL6 concentration was determined to be 348.5 pg/mL in unstimulated PBMC supernatant, 467.5 pg/mL in PHA stimulated PBMC supernatant.

Serum of sixteen individual healthy human donors was measured. The CXCL6 concentration of detected samples was determined to be 593.4 pg/mL with a range of 401.6-827.0 pg/mL.

Biolayer interferometry (BLl) kinetic assays of 85953-2-PBS against Human GCP-2/CXCL6 were performed. The affinity constant is 1.68 nM.
