The PLEKHG4 protein (full name: Pleckstrin Homology and RhoGEF Domain Containing G4), also known as Puratrophin-1, belongs to the Rho guanine nucleotide exchange factor (RhoGEF) family containing Dbl homology (DH) domains and pleckstrin homology (PH) domains. Its core function is to act as a GEF, catalyzing the conversion of small GTPases of the Rho family from the inactive GDP-bound state to the active GTP-bound state, thereby playing a key role in intracellular signal transduction and Golgi cytoskeletal dynamics.
