Background Information
Resistin (or 'resistance to insulin') was originally discovered in mice in 2001 and named for its ability to resist (interfere with) insulin action; at that time, it was proposed as a link between obesity and diabetes. Mouse resistin is an 11 kDa cysteine-rich polypeptide, and its gene is located on chromosome 8. It is synthesized as a 114 amino acid precursor, with a 20 aa signal sequence and a 94 aa mature segment. Neuroprotective nature of adipokine resistin in the early stages of focal cerebral ischemia in a stroke mouse model suggests that resistin may have a therapeutic potential effect in management of neurodegenerative disease.

1、我的样本稀释液有限,如何处理稀释倍数较大的样本?
我们建议您对样本进行梯度稀释。例如,人血清样本需要1000000倍稀释时,您可以分3个梯度,每个梯度100倍,进行稀释:
第一步:取2 uL人血清样本,加入198 uL样本稀释液,混匀得到100倍稀释的人血清样本
第二步:取2uL 100倍稀释的人血清样本+ 198 uL样本稀释液进行稀释,混匀得到10000倍稀释的人血清样本
第三步:取2uL 10000倍稀释的人血清样本+ 198 uL样本稀释液进行稀释,混匀得到1000000倍稀释的人血清样本。
如果样本需要进一步稀释,您可以使用更多梯度。除了我们推荐的样本稀释液,不建议您使用其他溶液或者水稀释。如果您的样本稀释液仍然不足,请联系我们的客服为您提供更多样本稀释液。
2、我没有恒温振荡器,这会对实验时间和结果产生影响吗?
如果您没有恒温振荡器,完成这个实验大约需要 2.5 小时。在加入抗体混合物后,我们建议您将板条37℃下静置孵育2小时。在加入 TMB 后,您只需让板条37℃下静置孵育15分钟即可。我们的研发人员通过对比实验已经证明,尽管信号会略有降低,但您仍然可以获得有效的实验数据。
