KE10161 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse IGFBP3 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse IGFBP3. The assay recognizes mouse IGFBP3. An antibody specific for mouse IGFBP3 has been pre-coated onto the microwells. The mouse IGFBP3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse IGFBP3 is added to detect the captured mouse IGFBP3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Insulin-like growth factor binding protein 3, IGFBP3, is an N-linked glycosylated, phosphorylated, secretory protein with known antiproliferative and pro-apoptotic functions. IGFBP3 belongs to a family of high affinity insulin-like growth factor (IGF) binding proteins, which function to sequester extracellular IGF-1, preventing IGF-1 activation of the insulin-like growth factor receptor, IGF-1R. IGFBP3 is highly expressed in the endometrium and at the maternal-fetal interface in multiple species, including humans. In a mouse embryo coculture system,IGFBP3 was found to be secretedby endometrial cells that promote embryonic developmentby enhancing the effect of IGF.
