KE10202 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse Fetuin A/AHSG ELISA kit is to be used to detect and quantify protein levels of endogenous mouse Fetuin A/AHSG. The assay recognizes mouse Fetuin A/AHSG. An antibody specific for mouse Fetuin A/AHSG has been pre-coated onto the microwells. The mouse Fetuin A/AHSG protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse Fetuin A/AHSG is added to detect the captured mouse Fetuin A/AHSG protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Fetuin-A, also named Alpha2-Heremans-Schmid glycoprotein (AHSG), is a 52 kDa glycoprotein synthesized in hepatocytes and secreted into the bloodstream: it is therefore addressed as a hepatokine. It acts as systemic inhibitor of extraosseous calcification, serves as transport protein for calcium and phosphate in colloidal calciprotein particles and is involved in bone mineralization. Fetuin-A is its interaction with the insulin receptor. Fetuin-A has also been identified as a biomarker for neurodegenerative disease. Fetuin-A modulation is associated with a number of pathophysiological variables that cause liver problems, including insulin receptor signaling deficiencies, adipocyte dysfunction, hepatic inflammation, fibrosis, triacylglycerol production, macrophage invasion, and TLR4 activation.
