KE00612 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Synaptotagmin-1 ELISA kit is to be used to detect and quantify protein levels of endogenous human Synaptotagmin-1. The assay recognizes human Synaptotagmin-1. An antibody specific for human Synaptotagmin-1 has been pre-coated onto the microwells. The human Synaptotagmin-1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Synaptotagmin-1 is added to detect the captured human Synaptotagmin-1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
The synaptotagmins are integral membrane proteins of synaptic vesicles thought to serve as Ca(2+) sensors in the process of vesicular trafficking and exocytosis. Synaptotagmin-1 acts as the primary calcium sensor for fast synchronous neurotransmitter release. It contains two C2 domains (C2A and C2B) that mediate Ca²⁺-dependent membrane interactions. Calcium binding to synaptotagmin-1 participates in triggering neurotransmitter release at the synapse. In addition to regulating exocytosis, synaptotagmin-1 has also been implicated in endocytosis and neurite outgrowth.
