KE00678 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse Desmin ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse Desmin. The assay recognizes human/mouse Desmin. An antibody specific for human/mouse Desmin has been pre-coated onto the microwells. The human/mouse Desmin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse Desmin is added to detect the captured human/mouse Desmin protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Desmin is the main intermediate filament protein in skeletal and cardiac muscle cells and is essential for the structural integrity and survival of muscle cells. As an abundant muscle-specific protein, desmin has been widely used as a marker of muscle derived tumors. Mutations in Desmin have been associated with Myofibrillar Myopathy 1, familial cardiac and skeletal myopathy (CSM). Anti-desmin is also valuable in the differential diagnosis of tumors of uncertain origin.
