Product Information

KE00708 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human KLK3/PSA ELISA kit is to be used to detect and quantify protein levels of endogenous human KLK3/PSA. The assay recognizes human KLK3/PSA. An antibody specific for human KLK3/PSA has been pre-coated onto the microwells. The human KLK3/PSA protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human KLK3/PSA is added to detect the captured human KLK3/PSA protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

KLK3 also known as Prostate-Specific Antigen (PSA), is a 33 kDa glycoprotein primarily synthesized by the epithelial cells of the prostate gland. Its main physiological function is to cleave seminogelins I and II in semen, leading to the liquefaction of the seminal coagulum and the release of motile sperm. It is under tight regulation by androgens via the androgen receptor (AR). Serum PSA levels are elevated in prostate cancer due to disruption of the glandular architecture, allowing more PSA to enter the bloodstream. It is used alongside digital rectal exam (DRE) for early detection. Although termed "prostate-specific," PSA is not exclusively produced by the prostate. Very low levels can be produced in periurethral glands, breast tissue, and some other tissues. Its expression is highly abundant in prostatic tissue and seminal fluid.
