KE00943 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human KAP1 ELISA kit is to be used to detect and quantify protein levels of endogenous human KAP1. The assay recognizes human KAP1. An antibody specific for human KAP1 has been pre-coated onto the microwells. The human KAP1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human KAP1 is added to detect the captured human KAP1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
KAP1, also named as TRIM28 or RNF96, is a 835 amino acid protein, which contain one RING-type zinc finger, one PHD-type zinc finger, one bromo domain and two B box-type zinc fingers. KAP1 localizes in the nucleus and Belongs to the TRIM/RBCC family. KAP1 is a nuclear corepressor for KRAB domain-containing zinc finger proteins and mediates gene silencing by recruiting CHD3, a subunit of the nucleosome remodeling and deacetylation (NuRD) complex, and SETDB1 to the promoter regions of KRAB target genes. KAP1 is a ubiquitously expressed protein including spleen, thymus, prostate, testis, ovary, small intestine, colon and peripheral blood leukocytes.
