KE00118 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The IL-6R ELISA kit is to be used to detect and quantify protein levels of endogenous IL-6R. The IL-6R ELISA kit is designed to measure human soluble IL-6R in serum, plasma, urine and cell culture supernatant. The assay recognizes human IL-6R. An antibody specific for IL-6R has been pre-coated onto the microwells. The IL-6R protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for IL-6R is added to detect the captured IL-6R protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Interleukin-6 (IL-6) is a pleiotropic cytokine produced by a variety of cells during infection, trauma, and immunological challenge. IL-6 acts via a receptor complex consisting of two distinct membrane-bound glycoproteins, an 80-kDa IL-6-binding subunit (IL-6R, CD126, gp80) and a 130-kDa signal-transducing element (gp130, CD130). After binding of IL-6 to membrane-bound IL-6R, the complex of IL-6 and IL-6R associates with gp130, thus activating the receptor. Expression of gp130 is found in almost all organs while expression of IL-6R is predominantly confined to hepatocytes and leukocyte subpopulations (monocytes, neutrophils, T cells, and B cells). Soluble form of IL-6R has been found in human serum and urine.

我们建议您对样本进行梯度稀释。例如,人血清样本需要1000000倍稀释时,您可以分3个梯度,每个梯度100倍,进行稀释:
第一步:取2 uL人血清样本,加入198 uL样本稀释液,混匀得到100倍稀释的人血清样本
第二步:取2uL 100倍稀释的人血清样本+ 198 uL样本稀释液进行稀释,混匀得到10000倍稀释的人血清样本
第三步:取2uL 10000倍稀释的人血清样本+ 198 uL样本稀释液进行稀释,混匀得到1000000倍稀释的人血清样本。
