KE00444 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human IL3RA/CD123 ELISA kit is to be used to detect and quantify protein levels of endogenous human IL3RA/CD123. The assay recognizes human IL3RA/CD123. An antibody specific for human IL3RA/CD123 has been pre-coated onto the microwells. The human IL3RA/CD123 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human IL3RA/CD123 is added to detect the captured human IL3RA/CD123 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Interleukin-3 receptor subunit alpha (IL3RA), also known as CD123, belongs to the cytokine receptor family. It is a glycoprotein containing an extracellular domain, involving a predicted Ig-like domain, two FnIII domains, a transmembrane domain and an intracellular domain. IL3RA is expressed on hematopoietic progenitor cells, plasmacytoid dendritic cells, basophils, monocytes, some B cells, and in a variety of hematologic malignancies. IL-3 initially binds to IL3RA and subsequently recruits the beta chain (CD131) to form the high-affinity receptor, resulting in the activation of a series of signaling pathways including the JAK/STAT, Ras-MAPK, and phosphatidylinositol 3-kinase pathways. IL-3R signaling regulates the proliferation, survival, and differentiation of hematopoietic cells.
