KE00784 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human HRAS ELISA kit is to be used to detect and quantify protein levels of endogenous human HRAS. The assay recognizes human HRAS. An antibody specific for human HRAS has been pre-coated onto the microwells. The human HRAS protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human HRAS is added to detect the captured human HRAS protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
HRAS (Harvey rat sarcoma viral oncogene homolog) belongs to the small GTPase superfamily and Ras family. RAS family proteins (KRAS4A, KRAS4B, NRAS and HRAS) function as GDP-GTP-regulated binary on-off switches, which regulate cytoplasmic signaling networks that control diverse normal cellular processes. HRAS is Involved in the activation of Ras protein signal transduction. As HRAS mutations are associated to activation of RAF/MEK/ERK signaling in different cancers. HRAS is mostly altered in head and neck squamous cell carcinoma (HNSCC) (5-9%), salivary glands (15%), and bladder cancer (5-30%).
