Product Information

KE00560 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human HER2/ErbB2 ELISA kit is to be used to detect and quantify protein levels of endogenous human HER2/ErbB2. The assay recognizes human HER2/ErbB2. An antibody specific for human HER2/ErbB2 has been pre-coated onto the microwells. The human HER2/ErbB2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human HER2/ErbB2 is added to detect the captured human HER2/ErbB2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

The human epidermal growth factor receptor (HER) family of receptors plays a central role in the pathogenesis of several human cancers. HER2 (human epidermal growth factor receptor 2), also known as ErbB2 and Neu, is 1 of the 4 membrane receptor tyrosine kinases (RTKs) and when activated affects cell proliferation and survival. HER2 is expressed in many tissues and its major role in these tissues is to facilitate excessive/uncontrolled cell growth and tumorigenesis. Additionally, HER2 mutations have also been reported in common cancers such as pulmonary, colorectal, and breast cancers, indicating a potential for HER2-directed therapies in these cancers.
