KE00609 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PSMA/FOLH1 ELISA kit is to be used to detect and quantify protein levels of endogenous human PSMA/FOLH1. The assay recognizes human PSMA/FOLH1. An antibody specific for human PSMA/FOLH1 has been pre-coated onto the microwells. The human PSMA/FOLH1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PSMA/FOLH1 is added to detect the captured human PSMA/FOLH1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
FOLH1 (folate hydrolase 1) is a type II transmembrane glycoprotein belonging to the M28 peptidase family. It is expressed in a variety of tissues, including the prostate, central and peripheral nervous system, and kidney. FOLH1 protein acts as a glutamate carboxypeptidase on different alternative substrates, including the nutrient folate and the neuropeptide N-acetyl-L-aspartyl-L-glutamate. In the nervous system, FOLH1 is involved in regulating the levels of the neurotransmitter NAAG and influencing neural signaling. In addition, FOLH1 is highly expressed in prostate cancer cells and is an important indicator of prostate cancer diagnosis and prognosis. The protein may also be associated with a variety of pathological conditions, such as glutamate excitotoxicity.
