KE00611 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human E-selectin / CD62E ELISA kit is to be used to detect and quantify protein levels of endogenous human E-selectin / CD62E. The assay recognizes human E-selectin / CD62E. An antibody specific for human E-selectin / CD62E has been pre-coated onto the microwells. The human E-selectin / CD62E protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human E-selectin / CD62E is added to detect the captured human E-selectin / CD62E protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
E-selectin (Endothelial Leukocyte Adhesion Molecule-1, also known as CD62E) is a critical adhesion molecule expressed on endothelial cells. It plays a crucial role in the initial steps of leukocyte recruitment to sites of inflammation, facilitating the rolling and adhesion of leukocytes to the endothelium. This process is essential for the immune response to infection and injury. E-selectin is upregulated in response to inflammatory cytokines such as IL-1, TNF-α, and bacterial lipopolysaccharide (LPS), and it interacts with various glycan ligands on leukocytes, including PSGL-1, ESL-1, CD44, and CD43.
