KE00818 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human E-cadherin ELISA kit is to be used to detect and quantify protein levels of endogenous human E-cadherin. The assay recognizes human E-cadherin. An antibody specific for human E-cadherin has been pre-coated onto the microwells. The human E-cadherin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human E-cadherin is added to detect the captured human E-cadherin protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
E-cadherin is a transmembrane glycoprotein that connects epithelial cells at adherens junctions. In normal cells, E-cadherin exerts its tumor-suppressing role mainly by sequestering β-catenin from its binding to LEF (Lymphoid enhancer factor)/TCF (T cell factor), which serves the function of transcribing genes of the proliferative Wnt signaling pathway. The loss of E-cadherin expression associated with the epithelial-mesenchymal transition (EMT) occurs frequently during tumor metastasis. Furthermore, Soluble E-cadherin (an 80-kDa soluble form) may be a biomarker for the diagnosis and prognosis of cancer.
