KE00728 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Cleaved Caspase 3/P17/P19 ELISA kit is to be used to detect and quantify protein levels of endogenous human Cleaved Caspase 3/P17/P19. The assay recognizes human Cleaved Caspase 3/P17/P19. An antibody specific for human Cleaved Caspase 3/P17/P19 has been pre-coated onto the microwells. The human Cleaved Caspase 3/P17/P19 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Cleaved Caspase 3/P17/P19 is added to detect the captured human Cleaved Caspase 3/P17/P19 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Caspases, a family of endoproteases, are critical players in cell regulatory networks controlling inflammation and cell death. Initiator caspases (caspase-2, -8, -9, -10, -11, and -12) cleave and activate downstream effector caspases (caspase-3, -6, and -7), which in turn execute apoptosis by cleaving targeted cellular proteins. Caspase 3 plays a key role in the activation of sterol regulatory element binding proteins (SREBPs) between the basic helix-loop-helix leucine zipper domain and the membrane attachment domain. Caspase 3 can form heterocomplex with other proteins.
