KE00421 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Cathepsin D ELISA kit is to be used to detect and quantify protein levels of endogenous human Cathepsin D. The assay recognizes human Cathepsin D. An antibody specific for human Cathepsin D has been pre-coated onto the microwells. The human Cathepsin D protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Cathepsin D is added to detect the captured human Cathepsin D protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CTSD (Cathepsin D) also named CPSD, belongs to the peptidase A1 family, which is synthesized in rough endoplasmic reticulum as preprocathepsin D. After removal of signal peptide, the 52 kDa procathepsin D is targeted to intracellular vesicular structures (lysosomes, endosomes, phagosomes). It is ubiquitously expressed and is involved in proteolytic degradation, cell invasion, and apoptosis. Loss of CTSD leads to lysosomal dysfunction and accumulation of different cellular proteins implicated in neurodegenerative diseases.
