KE00465 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CSPG4 ELISA kit is to be used to detect and quantify protein levels of endogenous human CSPG4. The assay recognizes human CSPG4. An antibody specific for human CSPG4 has been pre-coated onto the microwells. The human CSPG4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CSPG4 is added to detect the captured human CSPG4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CSPG4, also named as HMW-MAA, MCSP, MCSPG, MEL-CSPG, MSK16 and NG2, is a proteoglycan playing a role in cell proliferation and migration which stimulates endothelial cells motility during microvascular morphogenesis. CSPG4 may inhibit neurite outgrowth and growth cone collapse during axon regeneration. It is a cell surface receptor for collagen alpha 2(VI) which may confer cells ability to migrate on that substrate. CSPG4 may regulate MPP16-dependent degradation and invasion of type I collagen participating in melanoma cell invasion properties. CSPG4 is a signal-transducing protein by binding through its cytoplasmic C-terminus scaffolding and signaling proteins.
