KE00737 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CD107a/LAMP1 ELISA kit is to be used to detect and quantify protein levels of endogenous human CD107a/LAMP1. The assay recognizes human CD107a/LAMP1. An antibody specific for human CD107a/LAMP1 has been pre-coated onto the microwells. The human CD107a/LAMP1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CD107a/LAMP1 is added to detect the captured human CD107a/LAMP1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Lysosome-associated membrane glycoprotein 1 (LAMP1, also known as CD107a) is a glycosylated type I transmembrane protein that is crucial in lysosomal function and immune cell degranulation. It is enriched in the lysosomal membrane and provides selectins with carbohydrate ligands. LAMP1 is also a marker of degranulation in lymphocytes, such as CD8+ and NK cells, indicating its role in immune responses. Defects in LAMP1 can lead to impaired degranulation and reduced cytotoxicity, which are associated with conditions such as hemophagocytic lymphohistiocytosis. Additionally, LAMP1 is involved in tumor cell differentiation and metastasis.
