KE00936 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CA2 ELISA kit is to be used to detect and quantify protein levels of endogenous human CA2. The assay recognizes human CA2. An antibody specific for human CA2 has been pre-coated onto the microwells. The human CA2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CA2 is added to detect the captured human CA2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CA2(carbonic anhydrase II ) is also called Car2, CA-II, and CAII and belongs to the alpha-carbonic anhydrase family. It reversibly hydrates CO2 in cellular ion transport and homeostasis. CA2 is essential for bone resorption and osteoclast differentiation. It regulates SLC9A1 activity via a phosphorylation-regulated CAII binding site in SLC9A1 tail. It also may be involved in brain development. Defects in CA2 cause osteopetrosis autosomal recessive type 3 (OPTB3).
