KE00601 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human BCMA/TNFRSF17 ELISA kit is to be used to detect and quantify protein levels of endogenous human BCMA/TNFRSF17. The assay recognizes human BCMA/TNFRSF17. An antibody specific for human BCMA/TNFRSF17 has been pre-coated onto the microwells. The human BCMA/TNFRSF17 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human BCMA/TNFRSF17 is added to detect the captured human BCMA/TNFRSF17 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
BCMA (B cell maturation antigen), also known as TNFRSF17, is a type III transmembrane glycoprotein and is a member of the TNF-receptor superfamily. This receptor is preferentially expressed in mature B lymphocytes and plasma cells, which may be important for B cell development and autoimmune response. BCMA has two agonist ligands: a proliferation-inducing ligand (APRIL) and BAFF. When BCMA binds to APRIL, it transmits signals of cell survival and proliferation; when BCMA binds to BAFF, it mediates the activation of NF-kappaB and MAPK8/JNK. It has been found that the overexpression and activation of BCMA are associated with multiple myeloma (MM) in preclinical models and humans, supporting its potential utility as a therapeutic target for MM.
