GRHL3 is a conserved grainy‑head‑like epithelial transcription factor critical for epidermal barrier formation, embryonic morphogenesis and wound‑healing responses. It governs keratinocyte differentiation and balances epithelial cell proliferation versus differentiation. GRHL3 exerts dual‑faceted roles in malignancies; it acts as a tumor suppressor in skin squamous‑cell carcinoma while promoting proliferation and metastasis in colorectal cancer.
