ACOX2 (Acyl-CoA Oxidase 2) is a peroxisomal enzyme that catalyzes the first and rate-limiting step of branched-chain fatty acid β-oxidation, specifically targeting very long-chain fatty acids (VLCFAs) and bile acid intermediates. It plays a critical role in lipid metabolism by generating H2O2 and acetyl-CoA, thereby linking peroxisomal fatty acid oxidation to cellular energy homeostasis and redox signaling. Dysregulation of ACOX2 has been implicated in metabolic disorders, including non-alcoholic fatty liver disease (NAFLD) and certain peroxisomal biogenesis disorders, highlighting its potential as a therapeutic target for metabolic and liver-related diseases.
