KE00873 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CEBP Alpha/CEBPA ELISA kit is to be used to detect and quantify protein levels of endogenous human CEBP Alpha/CEBPA. The assay recognizes human CEBP Alpha/CEBPA. An antibody specific for human CEBP Alpha/CEBPA has been pre-coated onto the microwells. The human CEBP Alpha/CEBPA protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CEBP Alpha/CEBPA is added to detect the captured human CEBP Alpha/CEBPA protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CEBPA and its isoforms play important roles in lineage determination and gene activation in a variety of cell types by activating transcription from lineage-specific promoters. CEBPA is a DNA-binding protein that recognizes two different motifs: the CCAAT homology common to many promoters and the enhanced core homology common to many enhancers. In hematopoiesis, C/EBPa is a key factor in driving the development of myeloid cells interacting with a variety of factors, including c-Myc, PU.1, and microRNAs. It can also form heterodimers with the related proteins CEBP-beta and CEBP-gamma. The encoded protein has been shown to bind to the promoter and modulate the expression of the gene encoding leptin which plays an important role in body weight homeostasis. CEBPA can interact with CDK2 and CDK4, thereby inhibiting these kinases and causing growth arrest in cultured cells. Several pathways have been implicated as the means by which CEBPA mediates cell cycle arrest and proliferation, including p21, cyclin-dependent kinases and the E2F complex via c-Myc.

1、我的样本稀释液有限，如何处理稀释倍数较大的样本？
我们建议您对样本进行梯度稀释。例如，人血清样本需要1000000倍稀释时，您可以分3个梯度，每个梯度100倍，进行稀释：
第一步：取2 uL人血清样本，加入198 uL样本稀释液，混匀得到100倍稀释的人血清样本
第二步：取2uL 100倍稀释的人血清样本+ 198 uL样本稀释液进行稀释，混匀得到10000倍稀释的人血清样本
第三步：取2uL 10000倍稀释的人血清样本+ 198 uL样本稀释液进行稀释，混匀得到1000000倍稀释的人血清样本。
