Sandwich ELISA standard curve of MP50006-1, IL-17F Monoclonal Matched Antibody Pair, PBS Only. 68666-1-PBS was coated to a plate as the capture antibody and incubated with serial dilutions of standard  HZ-1116. 68666-2-PBS was HRP conjugated as the detection antibody Range: 62.5-4000 pg/mL.

Serum of sixteen individual healthy human donors was measured. Fourteen measured less than the lowest human IL-17F standard, 62.5 pg/mL. Two samples read 46.2 pg/mL and 277.7 pg/mL.

Human peripheral blood mononuclear cells (PBMC) was cultured in RPMI 1640 supplemented with 10% fetal bovine serum, 50 μM β-mercaptoethanol, 2 mM L-glutamine, and 100 μg/mL of streptomycin sulfate. The cells were unstimulated or stimulated with 1 μg/mL CD3 for 4 days. An aliquot of the cell culture supernatant was removed, the mean IL-17F concentration was undetectable in unstimulated PBMC supernatant, 3,282.8 pg/mL in CD3 stimulated PBMC supernatant.
