Background Information

APEX1 (Apurinic/Apyrimidinic Endonuclease 1), also known as APE1, HAP1, or Ref-1, is a multifunctional DNA repair enzyme. This protein is renowned for its bifunctional properties: on one hand, APEX1 is a core component of the base excision repair (BER) pathway, initiating DNA damage repair by cleaving apurinic/apyrimidinic (AP) sites to maintain genomic stability. On the other hand, APEX1 possesses redox activity, capable of reductively activating various transcription factors (such as AP-1, NF-κB, p53, HIF-1α, etc.) to regulate gene expression. Furthermore, this protein is involved in ribonucleotide reductase regulation, RNA processing, and mitochondrial DNA repair. APEX1 is highly expressed in various tumors and is closely associated with tumor drug resistance, radioresistance, and poor prognosis, making it a potential therapeutic target and prognostic marker. Its functional deficiency is linked to neurodegenerative diseases, cancer susceptibility, and aging.
