体内活性

腹腔注射HP-β-CD能显著提高白血病小鼠模型的存活率。将HP-β-CD系统性给药于小鼠,未观察到明显不良反应[2]。

体外活性

细胞处理(2-hydroxypropyl)-β-cyclodextrin能够激活转录因子EB(溶酶体功能和自噬的主要调节器)的激活,并增强细胞的自噬清除能力[1]。HP-β-CD处理通过G2/M细胞周期停滞和凋亡显著抑制白血病细胞生长,这是由于细胞内胆固醇的减少。72小时暴露后,HP-β-CD的IC50值在3.86–10.09 mM范围内。HP-β-CD还显示针对表达T315I BCR-ABL突变(这种突变对大多数ABL酪氨酸激酶抑制剂有抗性)的CML细胞和具有白血病干细胞特征的适应低氧的CML细胞具有抗癌效果。此外,HP-β-CD抑制人初级AML和CML细胞的集落形成能力[2]。
