体内活性

方法:为检测体内抗炎活性,将 DMOG (8 mg/mouse) 腹腔注射给小鼠,2 h 后注射 LPS (10 mg/kg) 诱导休克。 结果:DMOG 预处理可以减弱 LPS 诱导的 NF-κB 通路的全身激活。此外,用 DMOG 处理的小鼠在 LPS 诱导的休克中的存活率显著增加。[3]

体外活性

方法:PC12 细胞在常氧 (21% O2) 用 DMOG (1 μM-2 mM) 处理 24 h,使用 MTT assay 检测细胞活力。 结果:DMOG 引起显著的细胞毒性和活细胞部分的减少。[1] 方法:人肌腱干细胞 hTSC 用 DMOG (0.01-1 mM) 处理 96 h,使用 Western Blot 检测靶点蛋白表达水平。 结果:对 HIF-1α 核定位的分析显示,DMOG 以剂量依赖的方式激活该因子。[2]

细胞实验

To analyze DNA synthesis as an index of cellular proliferation, VSMC are plated in 48-well plates (5,000 per square centimeter) in growth medium, incubated overnight, and serum-deprived (1% FCS) for 24 h. Replicate wells are then stored at −70°C for baseline (day 0) cell counts, and fresh medium with or without growth factors is added to the remaining wells, which are incubated 72-96 h in 20 or 5% O2. Days 0 and 3 or 4 cell counts are determined by lysing cells in a buffer containing a fluorescent dye, which has minimal fluorescence by itself but fluoresces when bound to DNA or RNA. Absolute cell numbers are calculated by comparing the fluorescence of specimens with that of a standard curve similarly prepared using a known number of cells. (Only for Reference)
