Angiotensin II is a major biologically active vasoconstrictor peptide that regulates blood pressure by binding to AT1R and AT2R receptors. It stimulates the sympathetic nervous system, promotes aldosterone synthesis, and enhances renal function. Additionally, it induces vascular smooth muscle cell proliferation and increases collagen synthesis, leading to vascular and myocardial hypertrophy and fibrosis. It also promotes apoptosis and endothelial capillary formation. Angiotensin II is commonly used to induce hypertension and cardiac hypertrophy animal models.

体内活性

方法:为在人类高血压小鼠模型中测定胰岛素作用,使用渗透微型泵将 Angiotensin II (1.1 mg/kg in 0.9% saline) 给药给 C57Bl/6J 小鼠,持续二-四周。 结果:Angiotensin II 治疗后血压升高。葡萄糖给药后,Angiotensin II 治疗的小鼠的血清胰岛素增加幅度更大。长期 Angiotensin II 治疗四周可增强小鼠葡萄糖刺激的胰岛素分泌。[3] 方法:为分析体内遗传毒性效应,使用渗透微型泵将 Angiotensin II (60 ng/kg/min-1 μg/kg/min) 给药给 C57Bl/6J 小鼠,持续四周。 结果:Angiotensin II 使 SBP 比对照组高达 38 mmHg,并对小鼠的肾功能产生不利影响。在心脏中,在最高给药剂量下,可以检测到活性氧形成的显著增加和双链断裂。在肾脏中,观察到超氧化物形成的剂量依赖性增加、双链断裂和 DNA 碱基修饰突变。[4]

体外活性

方法:人肝癌细胞 (HCC) HepG-2、SMMC-7721、MHCC97-H 用 Angiotensin II (1-1000 nM) 处理 24-72 h,使用 MTT 方法检测的细胞活力。 结果:Angiotensin II 诱导 HCC 细胞系以时间和浓度依赖的方式显示出更高的生长。[1] 方法:新生大鼠心肌细胞用 Angiotensin II (1 μmol/L) 处理 5 min-48 h,使用 ELISA 和 RT-qPCR 方法检测 HMGB1 和 IL-6 表达水平。 结果:Angiotensin II 增强心肌细胞中 HMGB1 和 IL-6 的表达水平。[2]

动物实验

(129×C57BL/6) F1 mice, which lack AT1A receptors Angiotensin II used, are fed 10 gm/day gelled 0.25% NaCl diet that contains all nutrients and water. After 28 days of Angiotensin II infusion, hearts are harvested, weighed fixed in formalin, sectioned, and stained with Masson trichrome.
