体外活性

PROTACs通过连接两个不同的配体(一个为E3泛素连接酶的配体,另一个为靶蛋白的配体)实现其功能,利用细胞内的泛素-蛋白酶体系统有选择性地降解靶蛋白[1]。如5-溴-2'-脱氧尿苷(BrdU)及EdU在DNA活跃合成期被嵌入DNA中,随后可通过使用荧光偶氮探针及铜催化的1,3-偶极环加成反应进行检测。包括EdU在内的改性核苷类在长时间添加至细胞中可能具有细胞毒性[2]。5-Ethynyl-2'-deoxyuridine(EdU)染色是一种快速、敏感且可复制的方法,用于研究中枢神经系统中的细胞增殖[3]。
