Purity of recombinant human Wnt3A was determined by SDS- polyacrylamide gel electrophoresis. The protein was resolved in an SDS- polyacrylamide gel in reducing and non-reducing conditions and stained using Coomassie blue
Recombinant human Wnt3A (HZ-1296) induces dose-dependent alkaline phosphatase production in MC3T3 E1 mouse preosteoblast cells using pNPP as chromogenic substrate. MC3T3 E1 cells were treated with increasing concentrations of recombinant Wnt3A for 72 hours. The EC50 was determined using a 4-parameter non-linear regression model. The EC50 range is 2-17 ng/mL.
Recombinant human Wnt3A (HZ-1296) induces dose-dependent luciferase production in a HEK293 TCF/LEF reporter cell line. Luciferase production was assessed by One-Step™ luciferase assay Kit. HEK293 TCF/LEF reporter cells were treated with increasing concentrations of recombinant Wnt3A for 6 hours. The EC50 was determined using a 4-Parameter non-linear regression model. The EC50 range is 25-125 ng/mL.
Proteintech Wnt3A (HZ-1296) demonstrates a 3-fold lower EC50 and greater induction of luciferase production than a leading competitor. Recombinant human Wnt3A induces dose-dependent luciferase production in a HEK293 TCF/LEF reporter cell line.
Human induced pluripotent cells(iPSCs) were differentiated to definitive endoderm using HumanKine growth factors (Activin A- HZ-1138, bFGF-HZ-1285, BMP-4- HZ-1045, and Wnt3A-HZ-1296). The differentiation was confirmed by detection of expression of FOXA2(Red) and downregulation of pluripotency marker NANOG(yellow), in immunofluorescence
